Effects of Deep-Sea Water on Inhibition of Metastatic Regulators Expression in Human Colorectal Adenocarcinomas by Chitosan Oligosaccharide

Abstract
In this study, we investigated the effect of deep-sea water (DSW) on expressions of chitosan oligosaccharide (COS-L)-mediated metastatic factors, involving cyclooxygenase (COX)-2, urokinase plasminogen activate (uPA), and uPA receptor (uPAR), in HT-29 human colorectal adenocarcinomas. In order to evaluate the effect of DSW, COS-L was dissolved in DSW of various hardness (200, 400, 800, and 1,500) and then the mixtures were treated to HT-29 human colorectal adenocarcinomas, Expressions of COX-2 mRNA and protein induced by tumor necrosis factor (TNF)-¥á were significantly inhibited by mixtures of COS-L (3mg/mL) and DSW with various hardness, In addition uPA mRNA expression induced by phorbol-12-myristate-13-acetate (TPA) was decreased by the mixtures in a hardness dependent manner, but not uPAR. These data suggest that mixtures of COS-L (3 mg/mL) and DSW should inhibit metastasis by regulating only uPA transcription. Consequently, DSW effectively enhanced inhibitory effect of COS-L mediated metastatic factors, COX-2, uPA, which are essential enzymes for growth and metastasis in colorectal adenocarcinomas.
